what assay do you want to do ? how do you want to analyse ? there are several ways to do an enzyme assay.
one way to analyse how many units there are in your solution is to do a spectrometric analysis and measure the change in absorbance over time.
For example: ADH uses NADH when we add pyruvate. Thus by measuring the absorbance of NADH over time, we can calculate the concentrationchange over time and thus the amount of mols used per minute.

An assay is a procedure where the concentration of a component part of a mixture is determined.

So, an enzyme assay determines the concentration of an enzyme. There are lots of ways to do this. One is to put in the enzyme's substrate and see how quickly it reacts. Assuming you know the relationship between enzyme concentration and reaction rate you can find the answer.
Can you give more information about why you need to know this? We need more specifics to answer you.

dear frens,
let say the enzyme is polyketide synthase and i want to do an enzyme assay with the wild type and also its mutants. previous user said there are many ways to do it, so may i know for this specific enzyme, what is the best method for enzyme assay?
